<p>“Every cell, no matter from which organism, is always exposed to harmful substances in their environment that they have to deal with all the time,” said Gustavo Silva, assistant professor of biology at Duke and senior author on the paper. “Many human diseases are caused by cells not being able to cope with these aggressions.”</p>
<p>During the stress response, cells press pause the genes related to their normal housekeeping activities, and turn on genes related to crisis mode. Just like in a house being flooded, they put down the window cleaner, turn off the TV, and run to close the windows, then they patch holes, turn on the sump pump, and if needed, rip up carpet and throw away irreparably damaged furniture.</p>
<p>While studying mechanisms related to the cells’ health and their response to stress, the team saw that, under stress, a group of proteins was being modified inside the cells. They dug into it and found that the master regulator of this process is a gene called Rad6.</p>
<p>“When there is a stressor, cells need to change what proteins are produced,” said Vanessa Simões, associate in research in the Silva lab and lead author of the paper. “Rad6 goes in and gets the (protein-building) ribosomes to change their program and adapt what they are producing for the new stressful circumstances.”</p>
<p>Rad6 isn’t just any random gene. It can be found, sometimes under a different name, in almost all multicellular organisms. In humans, it is known for its association with a set of symptoms called “Nascimento Syndrome,” that include severe learning disabilities.</p>
<p>Nascimento Syndrome, also called X-linked intellectual disability type Nascimento, is still a poorly understood disease. It was officially described in 2006, and tends to run in families, giving scientists an early clue to its genetic causes. Affected individuals have severe learning disabilities, characteristic facial traits, with wide-set eyes and a depressed nose bridge, and a range of other debilitating symptoms.</p>
<p>Like many other genes, Rad6 doesn’t just do one thing. It’s a multiuse tool. By discovering an additional function, and one so tightly related to the cell’s health, Silva and his team get to add a new piece to the puzzle of Nascimento Syndrome.</p>
<p>“It’s still a big question or how exactly can a mutation to this gene lead to such a drastic syndrome in humans,” said Silva. “Our findings are exciting because Rad6 can be a model on which we can do genetic manipulations to try to understand how problems in coping with harmful conditions can be connected to how this disease progresses.”</p>
<p>“If we get a better understanding of how this gene works, we can actually try to interfere with it to help these patients have a better outcome.” he said.</p>
<p>But how does one actually “look” at what is happening with an infinitesimally small protein when a cell is stressed? With a fair amount of teamwork. Simões and Silva paired up with researchers from the Duke Biochemistry department and the Pratt School of engineering to gather all the help they needed.</p>
<p>“We used biochemistry analyses, cellular assays, proteomics, molecular modeling, cryo-electron microscopy, a whole set of advanced techniques,” said Silva.</p>
<p>“It’s the cool thing about being in a place like Duke,” he said. “We found collaborators and resources easily, right here, and that really increases the impact of a study and our ability to do a more complete work.”</p>

<p>As previously described, Maika Monroe stars as Julia. Julia moves to Romania with her husband, Francis. She doesn’t speak the language, she doesn’t know the city, she doesn’t know anyone. Julia notices a shaded figure standing, seemingly always staring, through the window across the street.</p>
<p>Monroe is no stranger to being stalked in cinema. Her breakout performance in <strong><em>It Follows</em></strong>  saw her endlessly pursued by a plodding supernatural being. After starring in <strong><em>The Guest</em></strong>,<em> It Follows</em>, <strong><em>Yes</em></strong>, <strong><em>Villains</em></strong>, <strong><em>The Stranger</em></strong>and <em>Watcher</em>her resume reads in part like a horror fan’s hand-picked list of stylish and scary stories. Collider’s own Perri Nemiroff spoke with Maika Monroe following <em>Watcher</em>’s premiere at Sundance, and she recapped her relationship with the horror genre,</p>

<p>Monroe’s versatility seems only matched by her passion, and both elements are on full display in her newest feature, <em>Watcher.</em></p>
<p><strong>Karl Glusman</strong> is Francis, Julia’s husband. Glusman carved out a career working with auteurs from across the world. He starred in <strong><em>Nocturnal Animals</em></strong>, <strong><em>Devs</em></strong>, <strong><em>Love</em></strong>,<strong><em>  Eternal Light</em></strong>and <strong><em>The Neon Demon</em></strong>. Glusman slides into the skin of his roles, sometimes to terrifying effect. While <em>Watcher </em>is the Maika Monroe show, Glusman always finds a way to make an impact with his contributions to the work.</p>
<p>Tiny spoiler ahead, so skip this paragraph if spoilers are of major concern, but the watcher himself is played by<strong> Burn Gorman</strong>. Gorman is a seasoned industry veteran with decades of experience, but he broke through to mainstream audiences with villainous performances in <strong><em>The Dark Rises</em></strong>  and <strong><em>Game of Thrones</em></strong>  as well as a charming turn as the straight-man across from <strong>Charlie Day </strong>in<strong><em>  Pacific Rim</em></strong>.</p>

<p>Despite in part being distributed by Shudder, <em>Watcher</em> won’t be released on the streaming platform alongside its theatrical debut. We already know <strong>which titles are coming to Shudder</strong> throughout June, and <em>Watcher </em>is nowhere to be seen–for now.</p>
<p><em>Watcher </em>was produced by Image Nation and Spooky Pictures, and it was acquired for distribution by IFC Midnight and Shudder. IFC started as the Independent Film Channel, but now the film distributor operates under two verticles: IFC Films and IFC Midnight. IFC Midnight is the genre film arm of the distributor responsible for fan favorites <strong><em>The Autopsy of Jane Doe</em></strong>, <strong><em>The Babadook</em></strong>and <strong><em>Baskin</em></strong>among many others. IFC President <strong>Arriana Bocco</strong> spoke in praise of director Chloe Okuno’s work in a <a target="_blank" href="https://www.amcnetworks.com/press-releases/ifc-midnight-and-shudder-takes-sundance-breakout-watcher-from-director-chloe-okuno/" rel="noopener noreferrer">press release</a> following the acquisition of the film saying,</p>

<p>As previously described, Maika Monroe stars as Julia. Julia moves to Romania with her husband, Francis. She doesn’t speak the language, she doesn’t know the city, she doesn’t know anyone. Julia notices a shaded figure standing, seemingly always staring, through the window across the street.</p>
<p>Monroe is no stranger to being stalked in cinema. Her breakout performance in <strong><em>It Follows</em></strong>  saw her endlessly pursued by a plodding supernatural being. After starring in <strong><em>The Guest</em></strong>,<em> It Follows</em>, <strong><em>Yes</em></strong>, <strong><em>Villains</em></strong>, <strong><em>The Stranger</em></strong>and <em>Watcher</em>her resume reads in part like a horror fan’s hand-picked list of stylish and scary stories. Collider’s own Perri Nemiroff spoke with Maika Monroe following <em>Watcher</em>’s premiere at Sundance, and she recapped her relationship with the horror genre,</p>

<p>I grew up loving old classic horror movies, watching <em>A Nightmare on Elm Street</em> and <em>The Shining</em>probably way too young, but there was just something about it. Unlike other movies, you always have such a visceral reaction to the genre. That was very intriguing to me. And then I ended up becoming an actor and ended up doing a movie called <em>It Follows</em>. That — not to gloat about it — but I think that film was the beginning of a change of the horror genre and kind of going back to its original roots of <em>The Shining</em> and [John] Carpenter. And so I think since that time there’s just been so many incredible genre films coming out, and so many people involved with the genre. So, for whatever reason, I just keep coming back! Especially with this script, I just fell in love with it.</p>

<p>Monroe’s versatility seems only matched by her passion, and both elements are on full display in her newest feature, <em>Watcher.</em></p>
<p><strong>Karl Glusman</strong> is Francis, Julia’s husband. Glusman carved out a career working with auteurs from across the world. He starred in <strong><em>Nocturnal Animals</em></strong>, <strong><em>Devs</em></strong>, <strong><em>Love</em></strong>,<strong><em>  Eternal Light</em></strong>and <strong><em>The Neon Demon</em></strong>. Glusman slides into the skin of his roles, sometimes to terrifying effect. While <em>Watcher </em>is the Maika Monroe show, Glusman always finds a way to make an impact with his contributions to the work.</p>
<p>Tiny spoiler ahead, so skip this paragraph if spoilers are of major concern, but the watcher himself is played by<strong> Burn Gorman</strong>. Gorman is a seasoned industry veteran with decades of experience, but he broke through to mainstream audiences with villainous performances in <strong><em>The Dark Rises</em></strong>  and <strong><em>Game of Thrones</em></strong>  as well as a charming turn as the straight-man across from <strong>Charlie Day </strong>in<strong><em>  Pacific Rim</em></strong>.</p>

These were brave bureaucratic organizations that were usually too far removed from practice.  The tricky point, however, is that the provinces are too often unable to distance themselves enough from interest groups.  I myself have seen up close that agrarian nature management has lost strength in terms of content as well as in terms of effectiveness.  I explicitly mention agricultural nature management here, because it is precisely the experiences of good nature management in agricultural areas that can provide a toolbox that is as practical as it is effective for the toolbox of Minister van der Wal.  As early as the 1970s, ecologists warned of impending problems with over-fertilization, linked to the loss of biodiversity.  There was no need for complicated calculation instruments.  Now that there is, the discussion gets bogged down in legal norms that are far removed from what works in practice. </p>
<p><b>Flanders as an example</b><br />I am regularly in Flanders for my work in the field of farmland birds.  The Belgians are very aware that the Flemish Land Agency (the DLG of Belgium) has a crucial role in the implementation of the nitrogen policy of the Flemish minister Zuhal Demir.  Demir knows that she can count on this government service and that the degree of influence by the Flemish counterparts of LTO Nederland, BoerenNatuur, Vogelbescherming and, for example, the World Wildlife Fund is relatively limited.  </p>

<p>MIT chemists have now designed a new type of photoredox catalyst that could make it easier to incorporate light-driven reactions into manufacturing processes. Unlike most existing photoredox catalysts, the new class of materials is insoluble, so it can be used over and over again. Such catalysts could be used to coat tubing and perform chemical transformations on reactants as they flow through the tube.</p>
<p>“Being able to recycle the catalyst is one of the biggest challenges to overcome in terms of being able to use photoredox catalysis in manufacturing. We hope that by being able to do flow chemistry with an immobilized catalyst, we can provide a new way to do photoredox catalysis on larger scales,” says Richard Liu, an MIT postdoc and the joint lead author of the new study.</p>
<p>The new catalysts, which can be tuned to perform many different types of reactions, could also be incorporated into other materials including textiles or particles.</p>
<p>Timothy Swager, the John D. MacArthur Professor of Chemistry at MIT, is the senior author of the paper, which appears today in Nature Communications. Sheng Guo, an MIT research scientist, and Shao-Xiong Lennon Luo, an MIT graduate student, are also authors of the paper.</p>
<p><strong>Hybrid materials</strong></p>
<p>Photoredox catalysts work by absorbing photons and then using that light energy to power a chemical reaction, analogous to how chlorophyll in plant cells absorbs energy from the sun and uses it to build sugar molecules.</p>
<p>Chemists have developed two main classes of photoredox catalysts, which are known as homogenous and heterogenous catalysts. Homogenous catalysts usually consist of organic dyes or light-absorbing metal complexes. These catalysts are easy to tune to perform a specific reaction, but the downside is that they dissolve in the solution where the reaction takes place. This means they can’t be easily removed and used again.</p>
<p>Heterogenous catalysts, on the other hand, are solid minerals or crystalline materials that form sheets or 3D structures. These materials do not dissolve, so they can be used more than once. However, these catalysts are more difficult to tune to achieve a desired reaction.</p>
<p>To combine the benefits of both of these types of catalysts, the researchers decided to embed the dyes that make up homogenous catalysts into a solid polymer. For this application, the researchers adapted a plastic-like polymer with tiny pores that they had previously developed for performing gas separations. In this study, the researchers demonstrated that they could incorporate about a dozen different homogenous catalysts into their new hybrid material, but they believe it could work more many more.</p>
<p>“These hybrid catalysts have the recyclability and durability of heterogeneous catalysts, but also the precise tunability of homogeneous catalysts,” Liu says. “You can incorporate the dye without losing its chemical activity, so, you can more or less pick from the tens of thousands of photoredox reactions that are already known and get an insoluble equivalent of the catalyst you need.”</p>
<p>The researchers found that incorporating the catalysts into polymers also helped them to become more efficient. One reason is that reactant molecules can be held in the polymer’s pores, ready to react. Additionally, light energy can easily travel along the polymer to find the waiting reactants.</p>
<p>“The new polymers bind molecules from solution and effectively preconcentrate them for reaction,” Swager says. “Also, the excited states can rapidly migrate throughout the polymer. The combined mobility of the excited state and partitioning of the reactants in the polymer make for faster and more efficient reactions than are possible in pure solution processes.”</p>

<p>The researchers also showed that they could tune the physical properties of the polymer backbone, including its thickness and porosity, based on what application they want to use the catalyst for.</p>
<p>As one example, they showed that they could make fluorinated polymers that would stick to fluorinated tubing, which is often used for continuous flow manufacturing. During this type of manufacturing, chemical reactants flow through a series of tubes while new ingredients are added, or other steps such as purification or separation are performed.</p>
<p>Currently, it is challenging to incorporate photoredox reactions into continuous flow processes because the catalysts are used up quickly, so they have to be continuously added to the solution. Incorporating the new MIT-designed catalysts into the tubing used for this kind of manufacturing could allow photoredox reactions to be performed during continuous flow. The tubing is clear, allowing light from an LED to reach the catalysts and activate them.</p>
<p>“The idea is to have the catalyst coating a tube, so you can flow your reaction through the tube while the catalyst stays put. In that way, you never get the catalyst ending up in the product, and you can also get a lot higher efficiency,” Liu says.</p>

<p>Flinders University researchers are focusing on how bacterial cells adapt and resist antimicrobial medications — with a new article focusing on a hospital strain of Acinetobacter baumannii and its cellular response to important antibiotic colistin.</p>
<p>The World Health Organization names antibiotic resistance as one of the biggest threats to global health, food security, and development with a growing number of infections — including pneumonia, tuberculosis, gonorrhoea and salmonellosis — becoming harder to treat as antibiotics used to treat them become less effective.</p>
<p>Antibiotic resistance leads to longer hospital stays, higher medical costs and increased mortality, researchers warn.</p>
<p>“Around the world, there are fewer and fewer new antibiotics being identified and produced for medical use — and this is compounded by the ever-increasing antibiotic resistance seen in bacterial strains causing infections,” says Flinders microbial researcher Dr Sarah Giles.</p>
<p>“If we can understand the bacterial mechanisms, such as this, we can potentially apply new therapies to treat patients — particularly those with multi-drug-resistant bacterial infections.”</p>
<p>As part of a NHMRC-Flinders University Research Scholarship study, Dr Giles and other authors noted that the A. baumannii bacterial strain had a two-part signal system which altered its potential response to antibiotic treatment.</p>
<p>This ‘two-component signal transduction’ observed involves a response regulator protein in the StkR/S system acting as a repressor and when genetically removed hundreds of transcriptional changes are seen.</p>
<p>Some of these changes affect the bacterial cell’s outer membrane composition leading to colistin resistance.</p>
<p>“Colistin is known as a ‘last resort’ antibiotic and therefore identifying and understanding the mechanisms contributing to bacterial antibiotic-resistant is critical,” says senior researcher Professor Melissa Brown.</p>
<p>Antimicrobial resistance (AMR) occurs when bacteria, viruses, fungi and parasites change over time and no longer respond to medicines, making infections harder to treat and increasing the risk of disease spread, severe illness and even death.</p>

<p>For those who got used to seeing David Harbour as Jim Hopper, a lazy police officer in Hawkins, Indiana, the new episodes of “Stranger Things” may catch you off guard — and that was his point. After three seasons of playing “fat dad Hopper,” Harbour was ready for something different.</p>
<p>The actor was excited to dive into the darker side of the character and how he was feeling about the trauma in his past. During Season 4’s fifth episode, he has a long monologue with the guard-turned-prisoner who tried to help him out and got caught. In the speech, during which Hopper details the pain of losing his daughter, he also says that he views himself as a “curse” and feels responsible for everything bad that has happened to El (Millie Bobby Brown) and Joyce (Winona Ryder). “Everyone I love, I hurt,” he says through tears.</p>
<p>As it turns out, Harbour had been sitting with that monologue for years and was eager to dive into Hopper’s backstory.</p>
<p>“I really liked the film noir idea of — it’s sort of in ‘Memento,’ where a man is chasing himself. We have this idea with Hopper: He’s a cop, so he sees things pretty much good guys, bad guys, pretty black and white. There’s a situation with cancer where his daughter dies. It’s cancer. What can you do, right? Who can you who can you lock up when a tragedy like this happen? Then what he really starts to do is go back, and the real bad guy is himself,” Harbour tells <em>Variety</em>. “I really liked the idea with him that there’s an actual poison in him. He is the poison, and feels that anyone who gets near him is infected with this thing. I feel like that’s why he had to lock himself away, it’s why he had to drink. To make him literally toxic, in a way, I thought was such a great metaphoric choice.”</p>
<p>After many conversations with Matt and Ross Duffer, Harbour began to mentally prepare.</p>
<p>“Then, lo and behold, it takes us six years to get to the actual shooting of it,” he says with a laugh. “They wrote that, and then I had two and a half years to sit there and, like, wait to do it. I read that scene two and a half years ago, and then COVID happened, then they started up with different stuff, and we didn’t shoot it until late in the season.”</p>
<p>Living with the speech for many years wasn’t easy for the actor: “I’m just so neurotic about it. You work on it, and then you don’t work — and you work on it again. You’re like, ‘What the fuck am I going to do with this thing?’ But I think it turned out pretty good. It was definitely a big part of my consciousness.”</p>

<p>In Episode 7, Hopper finally reunites with Joyce, a moment viewers were anxiously waiting for, as many hope to see a romance between the two. While it’s been revealed that the pair have somewhat of a history, it’s never been detailed — and Harbour isn’t sure whether that ever will be shared.</p>
<p>“It’s something that we’ve mapped out; how much they want to show is really up to them. Winona and I postulate a bunch of stuff, we have a bunch of theories. Some of them are very wild, and some of them are very tame,” he shares with a laugh. “In general, they had a time in high school — they knew each other when they were super innocent, and they had all this potential, both of them. Now they don’t have much potential. They’re sort of what they are. Knowing someone who, when you look in their eyes, you see that young person they were, is part of the bond between the two of them. They’ve known each other for so long and knew what you wanted to be and what you couldn’t be and what you couldn’t be. I think they have a very complex love for each other. It’s not just romantic. It’s much more complex than that.”</p>
<p>So, that reaches the question of whether it even is romantic at all — and if the duo could actually work out as a couple.</p>
<p>“I certainly feel like, from what happened in Season 3, it couldn’t work. The guy from Season 3 and woman from Season 3 could not work,” says Harbour. “Clearly there’s some chemistry and there is a longing for them to be together, but they would they would have to become different people.”</p>
<p>Ultimately, that choice is up to the Duffer Brothers: “What do they feel about characters that are ill-fitted to the world? Do they need to live happily ever after, or do they suffer in silence for the rest of their life? We’re not sure.”</p>

In the speech, during which Hopper details the pain of losing his daughter, he also says that he views himself as a “curse” and feels responsible for everything bad that has happened to El (Millie Bobby Brown) and Joyce (Winona Ryder). “Everyone I love, I hurt,” he says through tears.</p>
<p>As it turns out, Harbour had been sitting with that monologue for years and was eager to dive into Hopper’s backstory.</p>
<p>“I really liked the film noir idea of — it’s sort of in ‘Memento,’ where a man is chasing himself. We have this idea with Hopper: He’s a cop, so he sees things pretty much good guys, bad guys, pretty black and white. There’s a situation with cancer where his daughter dies. It’s cancer. What can you do, right? Who can you who can you lock up when a tragedy like this happen? Then what he really starts to do is go back, and the real bad guy is himself,” Harbour tells <em>Variety</em>. “I really liked the idea with him that there’s an actual poison in him. He is the poison, and feels that anyone who gets near him is infected with this thing. I feel like that’s why he had to lock himself away, it’s why he had to drink. To make him literally toxic, in a way, I thought was such a great metaphoric choice.”</p>
<p>After many conversations with Matt and Ross Duffer, Harbour began to mentally prepare.</p>
<p>“Then, lo and behold, it takes us six years to get to the actual shooting of it,” he says with a laugh. “They wrote that, and then I had two and a half years to sit there and, like, wait to do it. I read that scene two and a half years ago, and then COVID happened, then they started up with different stuff, and we didn’t shoot it until late in the season.”</p>
<p>Living with the speech for many years wasn’t easy for the actor: “I’m just so neurotic about it. You work on it, and then you don’t work — and you work on it again. You’re like, ‘What the fuck am I going to do with this thing?’ But I think it turned out pretty good. It was definitely a big part of my consciousness.”</p>

<p>In Episode 7, Hopper finally reunites with Joyce, a moment viewers were anxiously waiting for, as many hope to see a romance between the two. While it’s been revealed that the pair have somewhat of a history, it’s never been detailed — and Harbour isn’t sure whether that ever will be shared.</p>
<p>“It’s something that we’ve mapped out; how much they want to show is really up to them. Winona and I postulate a bunch of stuff, we have a bunch of theories. Some of them are very wild, and some of them are very tame,” he shares with a laugh. “In general, they had a time in high school — they knew each other when they were super innocent, and they had all this potential, both of them. Now they don’t have much potential. They’re sort of what they are. Knowing someone who, when you look in their eyes, you see that young person they were, is part of the bond between the two of them. They’ve known each other for so long and knew what you wanted to be and what you couldn’t be and what you couldn’t be. I think they have a very complex love for each other. It’s not just romantic. It’s much more complex than that.”</p>
<p>So, that reaches the question of whether it even is romantic at all — and if the duo could actually work out as a couple.</p>
<p>“I certainly feel like, from what happened in Season 3, it couldn’t work. The guy from Season 3 and woman from Season 3 could not work,” says Harbour. “Clearly there’s some chemistry and there is a longing for them to be together, but they would they would have to become different people.”</p>
<p>Ultimately, that choice is up to the Duffer Brothers: “What do they feel about characters that are ill-fitted to the world? Do they need to live happily ever after, or do they suffer in silence for the rest of their life? We’re not sure.”</p>
